In vitro study | Herniarin is cytotoxic to breast carcinoma cell line MCF-7 with an IC 50 of 207.6 µM. Herniarin (100 µM) also induces apoptosis in MCF-7 cells. Herniarin alone has no obvious cytotoxicity on transitional cell carcinoma (TCC) cells, but when in combination with 5 µg/mL cisplatin, Herniarin (80 µg/mL) potently enhances the antitumor effect of cisplatin, and increases chromatin condensation. |

fluorescence spectral properties | the three-dimensional fluorescence spectrum of 7-methoxycoumarin has two sets of fluorescence peaks, λex/λem = 215/392 nm (weak peak) and 325/392 nm (strong peak). Under the conditions of 7-methoxycoumarin strong acid, acidic to weak alkaline, weak alkaline to strong alkaline solution, the change trend of the fluorescence intensity of the solution is: enhanced, basically unchanged, weakened to almost no fluorescence. (2) When investigating the effect of different methanol-water ratio solvents on the fluorescence spectrum of 7-methoxycoumarin, it was found that with the increase of methanol volume content, the maximum emission wavelength of 7-methoxycoumarin had a blue shift, the fluorescence intensity had a downward trend, and the spectral shape basically did not change. (3) The fluorescence intensity of 7-methoxycoumarin was completely quenched by metal ion Fe3. Cu2 ,Ag and Ni2 reduced the fluorescence intensity of 7-methoxycoumarin slightly, while other metal ions had no effect. The standard curve of 7-methoxycoumarin was investigated, with good linear relationship and 0.999 correlation coefficient. (4) The fluorescence quantum yield of 7-methoxycoumarin was 0.44 at the excitation wavelength of 325nm by reference method, indicating that 7-methoxycoumarin is a strong fluorescent compound.
